Acetyl hexapeptide-8 is a synthetic chain of six amino acids, developed to interact with the proteins involved in releasing neurotransmitters at the junction between nerve and muscle.

How it works

Muscle contraction begins when a nerve releases acetylcholine into the gap between nerve ending and muscle. That release depends on a group of proteins called the SNARE complex, which draws the neurotransmitter-carrying vesicle to the cell membrane so its contents can be released.

Acetyl hexapeptide-8 is modelled on a segment of SNAP-25, one of the SNARE proteins. In laboratory studies it competes with the natural protein for its place in the complex, which is proposed to reduce the efficiency of vesicle release.

What is and is not established

Most of the mechanistic work has been done in cell culture and isolated tissue rather than on intact human skin. The open question is delivery: acetyl hexapeptide-8 is a water-soluble peptide, and the outer skin barrier is specifically structured to keep molecules of that type out. How much reaches the depth where the neuromuscular junction sits, from a topical product, is not well established.

Published human studies exist and report measurable changes, though many are small, manufacturer-supported, or use methods that make comparison across studies difficult.
