Carnosine is a dipeptide of two amino acids, beta-alanine and histidine, found naturally in human muscle and brain tissue at relatively high concentrations. It is one of the few cosmetic ingredients whose primary mechanism is neither antioxidant nor structural.

Glycation, which is the mechanism that matters

Glycation is a reaction in which sugars bind to proteins without enzymatic control, producing advanced glycation end products, usually abbreviated AGEs.

When this happens to collagen and elastin, the fibres become cross-linked and stiffer, losing the flexibility that lets skin recover its shape after movement. AGEs also carry a yellow-brown colour, contributing to sallowness.

This is a distinct route of skin ageing from UV damage and from oxidative stress, and most skincare ingredients do nothing about it.

How carnosine acts on it

Carnosine is described as a sacrificial target. It presents its own reactive sites to the sugars and reactive carbonyl compounds that would otherwise bind to collagen, effectively intercepting them.

Research also reports that carnosine may interfere with cross-links that have already formed, though the evidence for prevention is considerably stronger than for reversal.

Since glycation accumulates over decades and cross-links are not readily undone, prevention is the realistic framing.

Its other properties

Carnosine is also an antioxidant and a chelator of metal ions, particularly copper and zinc, which would otherwise catalyse oxidation reactions.

It has additionally been studied for buffering capacity, its principal role in muscle tissue, where it helps manage the acidity produced during exercise. That role is not relevant to topical use.

The evidence

Carnosine's biology is well characterised, with a substantial research literature spanning ageing, diabetes and exercise physiology. Glycation itself is thoroughly documented, particularly in diabetes research where AGEs are central to several complications.

The topical skincare evidence is thinner. Carnosine is a water-soluble dipeptide, and the barrier is structured to exclude such molecules, so the usual delivery question applies. It is small, which helps.

Why it appears alongside antioxidants

Because glycation and oxidation are different processes, an antioxidant does not address glycation and an anti-glycation agent does not scavenge free radicals in the same way.

Formulations aiming at comprehensive protection increasingly include both, which is why carnosine turns up in antioxidant serums rather than as a standalone product.

Related ingredients

Other anti-glycation ingredients include acetyl tetrapeptide-5, albizia julibrissin bark extract, and certain plant polyphenols. Each interferes with the process at a different point.

Diet, briefly

Carnosine is obtained from meat, and vegetarians have measurably lower tissue levels. That relates to ingestion and has no bearing on topical use, but it is a common question.
