Palmitoyl tripeptide-5 is a synthetic chain of three amino acids with a palmitic acid chain attached. It belongs to the same family as palmitoyl tripeptide-1, but its proposed mechanism is quite different.
TGF-beta, and why it matters
Transforming growth factor beta is a signalling protein central to how the body builds and repairs connective tissue. Among other things, it prompts fibroblasts to produce collagen.
TGF-beta is produced in an inactive form and must be activated before it can signal. That activation is regulated, which is one of the ways the body controls how much collagen gets made.
Palmitoyl tripeptide-5 is modelled on a sequence from thrombospondin-1, a protein involved in that activation step. The proposal is that it mimics thrombospondin's role, increasing the proportion of TGF-beta that becomes active, and so increasing the signal to produce collagen.
How that differs from other peptides
Matrikines such as palmitoyl tripeptide-1 are modelled on collagen fragments, signalling that damage has occurred. This one intervenes further upstream, in the regulation of a growth factor already present.
Neither adds anything to the skin. Both aim to influence what the skin does with what it has.
Why the palmitoyl chain
Peptides are water-soluble and the stratum corneum is structured to exclude water-soluble molecules. Attaching a palmitic acid chain improves lipophilicity and barrier compatibility.
It helps. It does not resolve the delivery question, which applies to every peptide in cosmetics.
The evidence
Research comes largely from the ingredient manufacturer, which is normal for cosmetic peptides, and independent replication is limited.
The underlying TGF-beta biology is well established from other fields. Whether a topically applied tripeptide meaningfully affects it in skin is the less settled part.
In formulation
Used at very low concentrations, stable, and typically combined with peptides acting through other routes rather than used alone.
